Pieris discovers and develops Anticalin® protein-based drugs to target validated disease pathways in a unique and transformative way. Our pipeline includes inhalable Anticalin® proteins to treat respiratory diseases and immuno-oncology multi-specifics tailored for the tumor microenvironment. Proprietary to Pieris, Anticalin® proteins are a novel class of therapeutics validated in the clinic and by partnerships with leading pharmaceutical companies. It is our mission to develop life-changing therapeutic proteins for patients suffering from cancer and respiratory diseases through the innovative application of novel science and cutting-edge technologies.

Your Tasks

  • Execute protein purification processes using ÄKTA FPLC systems from mg- to gram scale to ensure high quality material supply for pre-IND activities (clearance of host cell proteins (HCPs), DNA, endotoxin and other process and product related impurities)
  • Use in-process control methods including but not limited to cGE (SDS / IEF), SEC-HPLC, LAL, host cell-DNA, HCP ELISA to evaluate the quality of the material
  • Perform small-scale DoE experiments related to the establishment of new purification strategies using state-of-the-art and innovative chromatography and filtration techniques with an aim to setup lab scale platform DSP processes
  • Execute DSP development, optimization, and validation to support process up-scaling and transfer to CRO / CMOs
  • Serve as a technical leader for equipment and technologies for DSP development
  • Maintain accurate and detailed documentation of experimental methods and results using an electronic lab notebook
  • Compile and analyze experimental data to generate process knowledge and decide on next steps
  • Continuously optimize the process descriptions and their depiction in protocols and standard operation procedures (SOPs)
  • Assist in transfer of methods and processes to and from external labs

Our Requirements

  • Master or equivalent degree in Biotechnology, Biochemistry, Biochemical Engineering or related scientific area
  • Ideally more than two years of practical, ideally industrial experience in downstream processing
  • Hands-on experience with purification unit operations including filtration, chromatography, TFF, UF/DF and ÄKTA FPLC systems
  • Experienced in protein purification via affinity (AC), size exclusion (SEC), ion exchange (IEX), hydrophobic interaction (HIC) and/or mixed-mode chromatography (MMC)
  • Strong hands-on experience in troubleshooting and routine instrument maintenance
  • Knowledge and skills in biochemical characterization of proteins including analytical and biophysical methods
  • Knowledge in and experience with chromatographic resin screening, purification process development and process optimization and understanding of scale-up and GxP manufacturing is a plus
